Creative Biolabs Announces Unique Thioredoxin Library Construction Service
Shirley, August 19, 2015 (Newswire.com) - Thioredoxin is a class of small redox proteins known to be present in all organisms. It plays a role in many important biological processes, including redox signaling. In humans, it is encoded by the TXN gene. Loss-of-function mutation of either of the two human thioredoxin genes is lethal at the four-cell stage of the developing embryo. Although not entirely understood, thioredoxin plays a central role in humans and is increasingly linked to medicine through their response to reactive oxygen species (ROS). In plants, thioredoxins regulate a spectrum of critical functions, ranging from photosynthesis to growth, flowering and the development and germination of seeds. It has also recently been found to play a role in cell-to-cell communication. Because of its features, thioredoxin library construction has become an important technology for researchers.
Creative Biolabs' strong expertise in construction of library allows it to construct complex thioredoxin (TrxA) libraries without any dramatic increase in cost. Its thioredoxin library services include site-directed mutagenesis libraries, sequential permutation scanning libraries, and randomized and degenerated libraries.
TrxA scaffold represents a unique fashion of providing structural randomness among most popular protein scaffolds by replacing its active site loop with an inserted random peptide sequence. E. coli TrxA is originally a small cytosolic enzyme normally involved in maintaining the thiol or disulphide equilibrium inside the cell. It is highly soluble, rigid, and can be over expressed in large amounts. It was employed as a scaffold for the display of single conformationally constrained peptides. TrxA scaffold offers a reliable solution for potential application of short peptides which, by itself alone, is not applicable for its small size or other structural limitations.
The unique features of Creative Biolabs’ HiAffi™ platform include scalable production of libraries, controllable mutagenesis, applicable to high-throughput methods, large library capacity, fast turnaround time, and so on. The resulted thioredoxin library can be delivered in different formats, including but not limited to phage display library, ribosome display library, mRNA display library and cell surface display library.
